NKT cell abnormalities in EPTB and PTB patients. (A and C) Cocultures of APCs and T cells with α-GalCer. CD14⁺ monocytes and CD3⁺ T cells were purified from PBMCs by MACS. Monocytes were used as a source of APCs. Freshly isolated monocytes (1 × 10⁵ cells/well) were cocultured with T cells (9 × 10⁵ cells/well) for 7 days as described in Materials and Methods. Cells were then stained with PerCP-conjugated anti-CD3 and PE-conjugated anti-6B11 MAbs and analyzed by flow cytometry. The results shown are representative of the experiments. (B and D) Results were obtained from three independent donors for each group and are presented as means ± SDs.
