“In vivo” tracing of osteogenic-differentiated rMAPC cells after AAV6-GFP transduction in a calvaria bone CSD formation model. Macro-porous 3D hydroxyapatite-based scaffolds carried untransduced cells (a) or AAV6-GFP-transduced cells (b-e) and GFP expression was visualized by immunohistochemistry with anti-GFP antibody after 4 days (b) and 10 days (c) “in vivo” and with isotype IgG antibody after 4 (d) and 10 days (e). Sections were stained with DAB chromogen and counterstained with hematoxylin. Quantification of rMAPC-GFP+ cells “in vivo” (f) by light microscopy counting, mean from 5 technical replicates.
