The four main conformational states of the DAPY compounds based on the τ3 and τ4 torsional angles, illustrated for TMC278 . The U conformational state is centered around the “U”-shaped conformation with τ3 = τ4 = 0°. The U state corresponds to the bound conformation. The L1 and L2 conformational states are centered around the “L”-shaped conformations at (τ3,τ4) = (180°, 0) and (τ3,τ4) = (0°, 180°), respectively. Finally, the E or extended conformational state is centered around (τ3,τ4) = (180°, 180°)
