Mutating O-glycosylation sites or the FXYD motif inhibits surface expression of FXYD7. Oocyte were injected with 2 ng of cRNA coding for wild-type and mutated HA-tagged FXYD7 constructs ± the α1 (10 ng) and β1 (7 ng) subunits of Na+/K+ ATPase. Surface expression was measured 3 days later, and means ± S.E. of eight oocytes are depicted and expressed as a fraction of the mean value of oocytes expressing FXYD7.
