Effect of Gαs, Gαq/11, and Gα12/13 subfamily knockdown on GnRH-induced gonadotropin subunit transcription. LβT2 cells were transfected with control or Gα siRNA by nucleofection. Three days after siRNA transfection, cells were stimulated with either vehicle (no GnRH) or 1 nm GnRH for 2 h, followed by a 4-h incubation without GnRH, to induce gonadotropin subunit expression. The effect of Gαs, Gαq/11, and Gα12/13 knockdown on GnRH-induced gonadotropin subunit expression was determined by real-time PCR analysis.
