A, siRNAs and their targeted areas are depicted. B, SNF2L siRNA (12578) selectively knocked down SNF2L; SNF2LT siRNA (A1IS) selectively knocked down SNF2LT; and SNF2L/T siRNA (12667) knocked down both SNF2L as well as its isoform, SNF2LT. siRNA (NCSI) (RNAi-neg) was used as the negative control. ACTB served as housekeeping gene. C, singular knockdown of either SNF2L or SNF2LT significantly inhibited the growth of HM lines (MDA-MB-468 and MDA-MB-231). Dual knockdown of SNF2L as well as SNF2LT, however, was permissive of growth in HM lines. Each experiment was performed in triplicate and repeated at least four times.
