The effects of mutations in the nuclear localization signal of hairy/enhancer-of-split related with YRPW motif 1 (HEY1) or the variant L94M on the modulation of androgen receptor (AR)-mediated transactivation. U2OS cells were co-transfected with 100 ng of TAT-GRE-E1B-LUC, 25 ng of expression vector for AR and increasing amounts of expression vector for HEY1, L94M, the triple-point mutant HEY1-K51A/R53G/R51A or the triple-point mutant L94M-K51A/R53G/R51A (100 or 200 ng). The results shown represent the averages of the results of three independent experiments assayed in duplicate + s.d. *P<0.001 as compared with values for controls (Student’s t-test). **P<0.01 as compared with values for controls (Student’s t-test). After transfection, the cells were washed and incubated for 24 h in the presence of vehicle (white bars) or 10 nM DHT (black bars). Subsequently, cell lysates were assayed using a dual-luciferase reporter system. Data are presented as described for Figure 1b.
