(A) BMDC transfected with control or various MFG-E8 siRNAs (#1–#4) were loaded with apoptotic or necrotic cells prepared as described in Figure 2, and the protein levels of IL-1β, IL-6, IL-12p40 and IL-10 in culture supernatants were measured by ELISA. (B) BMDC from wild-type (WT) or MFG-E8-deficient mice (MFG-E8 KO) were loaded with dying cells as described above. In some instances, necrotic cells were treated with Necrostatin-1 for 48 h before coculture with BMDC. The protein levels of IFN-β, IL-6 and IL-12 were determined by ELISA.
