(A) Quantitative RT-PCR analysis of mRNA abundance of SREBP1a and SREBP1c in endometrial cancer cells. RNA abundance was shown as fold change relative to that in ECC-1 cells. (B) Western blotting (WB) analysis of lipogenic gene expression in commonly used endometrial cancer cell lines. Actin serves as protein loading control. (C) AN3CA cells were transduced with a set of lentiviral vector expressing shRNA targeting SREBF1. Western blot analysis of SREBP1 and SCD1 were performed showing a successful knockdown of SREBP1 and reduced expression of SCD1 as a transcriptional target of SREBP1. (D) Quantitative RT-PCR analysis of mRNA abundance of SREBP1, SREBP2 and their targets including FASN and SCD1 in #2 and #4 transduced cells.
