The presence of Sir2 helps forming higher order DNA–protein complex of PfORC1N1–238. EMSA was performed by incubating radio-labeled telomeric probe with PfSir2 followed by addition of increasing amount of PfORC1N1–238 with subsequent separation of DNA–protein complex in PAGE. Sir2 shows weak DNA binding (asterisk) (lanes 5 and 6) compared to ORC1N1–238 (lanes 2–4). Interestingly, supershift of ORC1-DNA band was observed in the presence of Sir2 (lanes 9–11). ‘Double asterisk’ indicates the supershifted bands relative to the black straight line.
