bcl-x deficiency does not cause compensatory changes in expression of other Bcl-2 proteins. (A) Primary embryonic fibroblast cells were isolated from 10 embryos of an 11-d-pregnant bcl-x heterozygous mouse. MEFs were immortalized by expressing SV40 T antigen and cloned. Bcl-xL expression in the established MEF cell lines was detected by Western blot analysis. (B) Changes in mRNA expression of Bcl-2 genes induced by bcl-x deficiency were determined by microarray analysis. Data are represented as fold changes in mean value of mRNA levels in two WT and two Bcl-x-KO cell lines (triplicate RNA samples for each cell line). Negative values indicate down-regulation. (C) Expression of Bcl-2 proteins in the indicated MEF cell lines was determined by Western blot analysis. KO #1, cell line 9; KO #2, cell line 4; WT #1, cell line 3; WT #2, cell line 6.
