We used 3-(4,5-dimethylthiahiazol-2-y1)-2,5-diphenyltetrazolium bromide and bromodeoxyuridine staining assays to determine that effects of heparanase expression on human retinal vascular endothelial cell proliferation. A: 3-(4,5-dimethylthiahiazol-2-y1)-2,5-diphenyltetrazolium bromide (MTT) assays revealed that cell proliferation in the heparanase small interfering RNA recombinant lentiviral vector (HPSE-LV)-Glu30 group was significantly suppressed by HPSE-LV compared with the Con-LV-Glu30 and Glu30 groups. Human retinal vascular endothelial cell (HREC) proliferation in the Glu30 group was significantly higher than in the Glu5 and HPSE-LV-Glu30 groups (●p<0.01 versus Glu5; ▲p<0.01 versus HPSE-LV-Glu30). Each group experiment was repeated 3 times (n=3). B, C: Bromodeoxyuridine (BrdU) staining assays revealed that HPSE-LV-Glu30 cell growth was significantly lower than in the Con-LV-Glu30 and Glu30 groups. The HREC growth of the Glu30 group was significantly higher than in the Glu5 and HPSE-LV-Glu30 groups. (●p<0.01 versus Glu5; ▲p<0.01 versus HPSE-LV-Glu30). Data are presented as the mean±SD.
