CCN3 promotes the adhesion, invasion and migration of RCC cells. (A) Adhesion of RCC cells to ECM proteins. Cells were plated in wells coated with fibronectin (FN) and laminin (LN) according to the manual. After 90 min, adherent cells were stained with 0.1% crystal violet then washed by immersion in tap water and dried. Adhesion was measured as absorbance at 550 nm. (B) For the migration assay, cells were seeded in the upper well of a transwell chamber. Medium with 10% fetal bovine serum was added to the lower well as the source of chemoattractants. Cells migrated through the polycarbonate membrane and were counted following H&E staining. (C) Invasion assays were performed using Matrigel-coated transwell chambers. The cells were embedded in Matrigel in the upper well, and the number of cells invading Matrigel and migrating to the lower well were counted following H&E staining. The data are the mean ± SD of three independent experiments. *P<0.05 vs. 786-O parental cells.
