Analysis of adipose progenitor cells (APC) by flow cytometry. Identification of adipogenic cell population in adipose stroma from white and brown fat tissues of mice fed a ND or HFD. Cells were initially selected by size, on the basis of forward scatter (FSC) and side scatter (SSC). The live singlets were sorted for lack of CD31 and Ter119 expression, and were further separated on the basis of expression of CD45. CD45 cells (Lin) were then sorted on the expression of CD34 and CD29. CD34+ cells were sorted on the basis of staining for Sca-1 and CD24 (right-hand plot). APC population changes in response to a HFD by flow cytometry in brown adipose tissue (A, B) and white adipose tissue (C, D). n=4 per group. *p < 0.05 compared with the ND group.
