IFN-alpha induces IFNaR2 cleavage in a TACE-dependent manner. HEK293T cells were transiently transfected with plasmids encoding IFNaR2 plus either TACE (Lanes 1–2) or vector DNA (Lanes 3–4). Each culture was split after 5 h and one half received IFN-alpha (1,000 U/mL) for 18 h, as indicated. Lysates were prepared and immunoblotted with anti-HA. Filters were stripped and re-probed with anti-EF2. These immunoblots are representative of 2 replicates.
