Ras signaling pathways are hyperactivated in Kras G12D/+ cortex. Protein lysates were prepared from 12- to 18-week-old control and CamKII-Cre; LSL Kras G12D/+ (Kras G12D/+) cortices. Activation of multiple signaling pathways downstream of Kras was measured by Western blot. The averages of controls are arbitrarily set at 1. Results are presented as average + S.D. Asterisks indicate statistically significant changes. A–C, phosphorylated and total levels of Akt (A, n = 3), ERK (B, n = 3), and p85/70S6K (C, n = 4) proteins were measured by Western blot. Phosphorylated protein levels relative to their total protein levels in Kras G12D/+ cortices were quantified and compared with those in control cortices. D, expression levels of IκBβ, a negative regulator of NF-κB signaling, relative to β-actin levels in Kras G12D/+ cortices were quantified and compared with those in control cortices (n = 3).
