Effects of NOV-002 on the MAPK pathway. Concentration-dependent effects of NOV-002 on stress kinases and their phosphorylated products are consistent with actin glutathionylation patterns. HL60 cells were treated for 1 h with 0 to 500 μmol/L NOV-002 in complete medium. Fifty micrograms of protein lysate were separated by SDS-PAGE and analyzed by immunoblot for phosphorylated and total levels of (A) JNK, (B) p38, and (C) ERK. The corresponding relative abundance of phosphorylated proteins was plotted as the relative ratio to actin.
