HBZ binds the HAT domain of p300/CBP. (A) Schematic representation of CBP and the CBP fragments used in GST pull-down assays. (B) C-terminal truncation of the HAT domain disrupts HBZ-binding. HBZ was incubated with 10 pmol of GST, GST-HAT or GST-N-HAT. Proteins retained on glutathione beads were analyzed by SDS–PAGE and autoradiography. (C) HBZ interacts with a region of p300/CBP encompassing the C-terminal portion of the HAT domain and the ZZ domain, and separately with the TAZ-2 domain. HBZ was incubated with 10 pmol of the indicated GST fusion proteins. Proteins retained on the glutathione beads were analyzed by SDS–PAGE and autoradiography.
