Analysis of BACE1 cleavage of CHL1 and L1 in COS cell cultures. A and B, COS cells were transiently co-transfected with CHL1-V5His (or L1) and BACE1, and the growth media were replaced with fresh medium and conditioned for 24 h with or without 10 μm β-secretase inhibitor IV (BI IV). Cell lysates were analyzed by Western blot for full-length and C-terminal fragments of CHL1-V5His and L1 using an anti-V5 mAb and an anti-L1(C-20) pAb, respectively. The soluble fragments of CHL1 and L1 were detected from the conditioned medium using an anti-CHL1(N-14) pAb and an anti-L1(N-14) pAb, respectively. C, semi-quantification of the Western blots for the secreted (soluble) CHL1 and L1 levels. Data were mean ± S.E., n = 4, Student's t test (*, p < 0.05; ns, statistically not significant).
