The Genetic Strategy to Isolate Mutants Defective in the Uniparental Inheritance of the cpDNA.

(A) Mutants defective in the uniparental inheritance of the cpDNA were obtained through insertional mutagenesis of mt+ cells, followed by crossing the transformants with a tester strain carrying a spectinomycin resistance marker gene (aadA) in its cpDNA (CL218 mt−).

(B) The bp31 mutant showed a higher rate of transmission of mt− DNA (aadA) than the control.
