(A) Colorectal cancer cells DLD1 and HCT116 were stably transfected with a doxycycline (Dox)-inducible expression vector encoding the DRO1 protein to yield DLD1/DRO1 and HCT116/DRO1 cells. Cells were stimulated with Dox for the indicated periods of time. Cells were harvested and lysates were analyzed for DRO1 expression by immunoblotting. (B) DLD1/DRO1 and (C) HCT116/DRO1 cells were stimulated with Dox (or left untreated) and treated with increasing amounts of the CD95-activating antibody APO. Cells were stained using propidium iodide and the number of apoptotic cells was determined using flow cytometry.
