The depletion of MINK1 induces multinucleated cells. A, HeLa cells were cultured on glass coverslips and transfected with the indicated siRNAs. Seventy-two hours later, cells were fixed and immunostained with anti-α-tubulin antibody and Hoechst. The arrows indicate multinucleated cells (scale bar, 50 μm). B, HeLa cells that constitutively expressed GFP-tagged siRNA-resistant wild-type or kinase inactive MINK1 were established by retroviral infection. The cells were treated as in A, and the ratio of multinucleated cells was evaluated. Three independent experiments were performed, and 200 cells were evaluated for each experiment (the data are represented as the mean ± S.D.; **, p < 0.01; n.s., not significant). C, cells were transfected with the indicated siRNA; 72 h later, the cells were lysed and immunoblotted with anti-MINK1 antibody.
