HPPL-Grhl2 starts expanding the lumen without proliferation. (A) Cysts derived from HPPL-Grhl2 contain more cells than the control at day 7 of 3D culture. Cysts were fixed in 4% PFA, and nuclei were stained with Hoechst 34580. Pictures were taken on a microscope, and the number of cells surrounding the lumen was counted. (B) Cysts derived from HPPL-Grhl2 contain similar number of cells at day 4 of 3D culture. (C) HPPL-Grhl2 expands the lumen without changing the number of cells forming the lumen. Images of cysts derived from the control HPPL and HPPL-Grhl2 at different time points were taken under a confocal microscope. The numbers of cells around the lumen are indicated under the image at each time point. Bars, 50 μm.
