(A) A read can simply be another known miRNA with an A:G mismatch to the source. (B) Read R1 is an HP-read that has a C:T mismatch with read R2. Read R2 looks like the edited version of miR-421 with an A:G mismatch in position 4. (C) Shows a read, R, with a negative hairpin test (NHP-read), comprising only one mismatch with any known miRNA, M. The closest read with a positive hairpin to this read has more than K mismatches with it, thus the read R passes the K-safety filter.
