A) Hep-3B cells were transfected with control (siRNA-Ct), silencer gp96 (siRNA-gp96) or nontransfected (φ) for 18 h. Gp96 expression was analyzed by western-blot using anti-Gp96 antibodies (α-Gp96) followed by signal quantification with ImageJ software. Right panel shows the levels of fibronectin expression of transfected cells. Adhesion of S. aureus V329 wild type (Wt) and Bap deficient strain (Δbap) on Hep-3B cells transfected with control (siRNA-Ct) or silencer gp96 (siRNA-gp96) (B) or to Vero and GPC-16 cells that do not express Gp96 at the cell surface (C). After 1 h of infection, cells were lysed and cell extracts were vigorously vortexed for 2 min. Bacterial adhesion was measured by CFU counts. Error bars represent the standard deviation of at least 3 independent experiments.
