(A) Whole human pulmonary artery segments were cultured for 48 hours in the presence of media alone (CTRL), C12-iE-DAP 1 µg/ml (NOD1 agonist) or LPS 1 µg/ml (TLR4 agonist). CXCL8 release was used as a measure of vessel activation. Results are expressed as the percentage (control ± SEM) of CXCL8 release under basal conditions (CTRL) for n = 6 donors. Results were analysed by one-sample t-test (*P<0.05). (B) Human lung microvascular endothelial cells (HMVEC) and (C) Human pulmonary artery smooth muscle cells (HPASMC) were cultured for 24 hours in 96 well plates in the presence of media alone, C12-iE-DAP 1 µg/ml (NOD1) or LPS 1 µg/ml (TLR4). Results are mean ± SEM for n = 12 (HMVEC) or n = 6 (HPASMC). Results were analysed using 1-way ANOVA with Bonferroni’s Multiple Comparison Test. (*P<0.05 vs. CTRL), (+P<0.05 vs. LPS).
