Reaction mixtures (100 μl) containing 1 μg of protein (MTB-PPX1, Rv1026, E. coli GPP (EC-GPP) or E. coli PPX (EC-PPX)), 1 mM GTP, 1 mM MnCl2, 4 mM (NH4)2SO4, in 50 mM Tris-HCl buffer (50 mM, pH 7.4), were incubated at 37°C for 0 or 60 minutes. Product mixtures were directly analyzed using anion exchange chromatography. Chromatograms obtained are shown in Panels A–D. Panel A: MTB-PPX and GTP. Panel B: Rv1026 and GTP. Panel C: EC-GPP and GTP. Panel D: EC-PPX and GTP.
