Liver leukocytes were prepared from C57BL/6J (WT) or IL-10−/− mice that were infected with MCMV for 5 and 7 days. As described in Materials and Methods, cells were stimulated with M45 viral peptide, stained for surface expression of CD8α and TCRβ, fixed, permeabilized, and stained for intracellular IFN-γ or TNF-α. Results show the frequencies total numbers of CD8+ T cells expressing (A and B) IFN-γ or (C and D) TNF-α. Data are combined from three independent experiments (n = 9–10 mice per group for each time point tested) and show the means ± SE. Asterisks denote statistically significant differences between WT and IL-10−/− groups, where p values are ≤0.05 (Student's T test).
