Inhibitory effect of PEA-15-AA on ovarian cancer proliferation, migration capability and angiogenesis is dependent on β-catenin inhibition. (a) β-Catenin siRNA inhibited SKOV3.ip1 cell proliferation. After 48 h transfection with siRNA, cells were trypsinized, and 1 × 105 cells were plated. Total cell numbers were counted after 48 h. (b) β-Catenin siRNA inhibited SKOV3.ip1 cell projection formation. (c) Wnt/β-catenin inhibitor suppressed SKOV3.ip1 cell projection formation. Cells were pretreated with 1 μM of pyridium pamoate (Sigma-Aldrich) for 1 h, and 3D projection formation assay was performed. (d, e) Rescue of β-catenin expression in PEA-15-AA stable clones recovered projection formation ability (d) and colony formation ability (e). Cells were transiently transfected with β-catenin plasmid, and then 3D projection formation assay and colony formation assay were performed.
