Rapid heating and cooling reduces the formation of multiple conformations in a ssRNA Pentaprobe. ³²P-labeled PP3 was subjected to denaturation and rapid cooling and then analysed on a 4% polyacrylamide gel; for comparison, a non-heated probe was also analysed. Lane 1: probe PP3 pre-denaturation, showing multiple bands that most likely reflect different secondary structure or oligomerization states. Lane 2: PP3 post denaturation.
