The effects of MB on the surface expression of SERT in EM4 cells. EM4 cells were transfected with hSERT and assayed for the fraction of hSERT expressed at the plasma membrane in the absence and presence of 10 µM MB. Total specific binding as defined by subtracting the binding of [¹²⁵I]RTI-55 (20 nM) achieved in the presence of paroxetine (10 µM) alone from total binding and surface-specific binding as calculated by subtracting [¹²⁵I]RTI-55 binding in the presence of 100 µM 5-HT from that of [¹²⁵I]RTI-55 alone. Data are given as mean ± SEM from three separate experiments (n= 11–14). No significant difference between MB and the vehicle was detected.
