The activity of thrombin (10 pM) was assayed in the presence of different NaCl concentrations measuring the hydrolysis of the fluorogenic substrate Boc-Ile-Glu-Gly-Arg-7-amido-4-methylcoumarin. (a) Thrombin inhibition as function of cE5 concentration in the presence and absence of NaCl as indicated. (b) Comparison of thrombin inhibition by the An. gambiae cE5 and the An. albimanus anophelin in the absence of salt in the assay buffer and without pre-incubation. (c) Comparison of thrombin inhibition by the An. gambiae cE5 and the An. albimanus anophelin in the presence of different NaCl concentrations (25 mM, 50 mM, 100 mM, 150 mM) in the assay buffer. Thrombin (10 pM) was pre-incubated (10 min) with 1 nM cE5 or anophelin before addition of the fluorogenic substrate. Data represent the mean of triplicates. Error bars represent SEM. In (A) and (B) the activity of thrombin in the absence of inhibitor was assumed as 100%.
