Knockdown of CARP-1 blocks WA effects. Cells were transfected with 100 nM each of scrambled or CARP-1 siRNAs for 72 h and then were either untreated (Control, DMSO) or treated with 10 µM WA for further 24 h. Cell lysates were subjected to western blotting as in Fig. 4 for levels of CARP-1 and actin proteins or subjected to MTT assay for determination of cell viabilities essentially as in Fig. 1. Columns in B represent means of three independent experiments; bars, S.E. * and #, p = ,0.01 relative to WA-treated, scrambled siRNA-transfected cells.
