MS/MS verification of Asn-182 glycosylation. To localize the glycosylation of the hexose-modified VP4-derived peptide, ¹⁷¹TNSPDYSLAYSNTTEK¹⁸⁷, the MS/MS spectrum of the [M+2H]²⁺ base peak of this peptide, observed at m/z 1026.97, was acquired through mass-dependent acquisition by the LTQ Orbitrap XL mass spectrometer. The C-terminus-containing y-ion fragments, indicated by stars, allowed the localization of the hexose modification at the Asn-182 residue. The inset in the top panel shows the fragmentation scheme, with the bound hexose being represented by the full circle. Those regions of the spectrum magnified ×5 or ×50 are indicated. The lower panel shows an expansion of the region between m/z 840 and 860.
