Western blot analysis of the effects of the N-alkoxy I3C derivatives on CDK6 and CDK2 protein expression. MCF-7 cells were treated with 200 μM I3C, 10 μM N-methoxy I3C, 5 μM N-ethoxy I3C, 1 μM N-propoxy I3C, and 0.5 μM N-butoxy I3C, or with 100 μM tryptophol, and harvested at the indicated time points. Total cell extracts were electrophoretically fractionate in SDS polyacrylamide gels, and western blots were probed with specific antibodies to either CDK6 or CDK2. Equal gel loading was confirmed using Ponceau S staining of the western blot membrane.
