(A) The percentage of FITC+ MHCII+ skin cells migrating to inguinal DLNs of WT (Eps8+/+) or Eps8−/− (Eps8−/−) mice painted on the abdomen with 0.5% FITC in 1:1 acetone:dibutylphtalate mixture was assessed by flow cytometry. Numbers in the gate are the percentage of FITC+ MHCII+ positive cells (6 mice/group, two independent experiments).

(B) The absolute numbers of FITC+ MHCII+ CD11c+ cells migrating to the DLNs at different time points in WT (Eps8+/+) and Eps8-deficient (Eps8−/−) animals. Data represent the mean ± SD (6 mice/group, out of two independent experiments).

(C)FITC+MHCII+CD11c+ cells were analyzed for the expression of CD24 and CD207. Numbers in the quadrants show the percentage of positive cells.

(D) The absolute numbers of FITC+ dermal DCs (CD24 CD207), langerin dermal DCs (CD24 CD207+) and Langerhans cells (CD24+ CD207+) migrating to the DLNs at different time points in WT (Eps8+/+) and Eps8-deficient (Eps8−/−) animals.
