The ScTrl1 healing domain inhibits the rescue of PaOrf2 toxicity by Pnkp and Rnl1. Serial 10-fold dilutions of S. cerevisiae cells expressing PaOrf2 (PaT) from a CEN vector or an empty vector or in the presence of an additional 2μ plasmid encoding full-length ScTrl1 or its component end-healing (Kin-CPD) or end-sealing (LIG) domains were spotted on –Ura–His agar plates with 2% glucose or galactose. The plates were photographed after incubation at 30°C for 2 d (glucose) or 3 d (galactose).
