Immunoblotting of embryoid bodies transduced with Ad-LacZ or Ad-Myo confirm myocardin overexpression at a variety of developmental times (A). Immunofluorescent assessment of day 28 embryoid bodies reveals an increase in SMαA, calponin and SMMHC (B) staining following treatment with Ad-Myo from day 10 onwards, in comparison to no treatment or Ad-LacZ negative controls. Photomicrograph exposure times were kept constant across all three treatment groups and deliberately low enough to clearly show the large increases in fluorescent intensity in the Ad-Myo group.
