Cultures of E. coli MC4100 and a KLH11 derivative with in-frame deletions of ssaI and ssbI expressing plasmid-borne ssaI or ssbI were extracted and subjected to reverse-phase chromatographic separation prior to tandem MS analysis using the precursor ion-scanning mode (transitions were monitored for precursor [M + H]+ -> m/z 102) for (A) MC4100 + SsaI, (B) MC4100 + SsbI, (C). KLH11 ssaI–ssbI–, (D) KLH11 ssaI–ssbI + Plac-ssaI, (E) KLH11 ssaI–ssbI + Plac-ssbI. The peaks in the chromatograms are labeled with lowercase lettering and include species defined in Table 1 and the AHLs noted.
