Flagellins expressed in Hfx. volcanii Δagl strains other than the ΔaglD strain migrate faster than when expressed in the wild type. Western blot analysis was performed on protein extracts from Hfx. volcanii H53 (wild type) or Δagl strains grown to the mid-log phase in MGM medium and expressing either FlgA1-His (A) or FlgA2-His (B), encoded by genes expressed under the regulation of a trp-inducible promoter. His-tagged proteins were detected by using anti-His antibodies. Comparable amounts of protein were loaded into each lane of panel A as well as panel B. There was twice as much sample loaded for expression in panel B as in panel A, since FlgA1-His appears to be more abundant than FlgA2-His in the cell extracts. The migration of molecular mass standards is indicated on the left (in kDa).
