(A and F) Down regulation of PTEN and/or overexpression of HER2 in MCF7 or Sum159 cells is demonstrated by western blotting. (B and G) PTEN down regulation in HER2 overexpressing cells increased the tumorsphere formation, (C and H) Aldefluor-positive cell populations (D and I) as well as invasion compared to either HER2 overexpression or PTEN down regulation in vitro. (E) MCF7-HER2+PTEN cells showed a significant increase in the proportion of CD44+/CD24− cells as compared to the MCF7-HER2+ or MCF7PTEN cells.
