(A) HEK293T cells were co-transfected with plasmids encoding GFP-AMPK-α2 and Flag-SIRT1 for co-immunoprecipitation studies. B) Increasing amount of His-SIRT1 was incubated with GST or GST- AMPK-α2 (1-312) and the complex was pulled down with glutathione-Sepharose beads. The presence of proteins in the complex was detected with the indicated antibodies. C) Plasmids encoding GFP-AMPK-α2 and Flag-SIRT1 were co-transfected into HepG2 cells for co-immunofluorescence staining. Flag-SIRT1 was visualized by anti-Flag antibody and secondary antibody conjugated with Texas-Red. D) Similar to B) but His-tagged truncations of SIRT1 (i) M1-a.a.1-235; (ii) M2-a.a.236-525; (iii) M3-a.a.526-747 were incubated with GST or GST- AMPK-α2-CAD.
