(A) SPARC protein expression in 7 types of gastric cancer cell lines. (B) SPARC protein (upper panel) and mRNA (lower panel) expression in parental cells (BGC-P and HGC-P), empty vector transfected clones (BGC-EV and HGC-EV), SPARC cDNA–expressing clone (BGC-SP) and SPARC shRNA-expressing clone (HGC-sh). GAPDH served as loading control. (C) Cell proliferation was determined by MTT assay. BGC-P and HGC-P cells and transfected clones (500 cells) were cultured in 96-well plates and incubated for 8 days, and cell proliferation was assayed by MTT method. Results are shown as means (±s.d.) of quadruplicate determinations from six separate experiments.
