HA and Myc tags do not severely affect Mus81 or Mms4 protein levels. (A) Strains containing HA- or Myc-tagged Mus81 (WDHY2420 and WDHY1868, respectively) or Mms4 (WDHY2423 and WDHY1872, respectively) were grown to an OD600 of 1, and protein was extracted by bead disruption and trichloroacetic acid precipitation. Fifty micrograms of total lysate was separated by electrophoresis, transferred to nitrocellulose, and subsequently immunoblotted with anti-Mus81, anti-Mms4, anti-HA, or anti-Myc antibodies to determine steady-state protein levels. Probing with anti-3-phosphoglycerate kinase (3-PGK) antibodies served as a loading control. (B) Quantification of Mus81 protein signal. (C) Quantification of Mms4 protein signal.
