^*WNV isolates COL524/08 and COL9835/08 were characterized by their temperature sensitivity (TS) phenotypes in Vero and BHK₂₁ cells. Temperature sensitivity assays were conducted at 37 and 39.5°C. Cells were seeded in 6-well plates and infected in duplicates at MOI of 0.01. At 24–72 h intervals post-infection, cells, and media from each well were harvested and centrifuged at 3,000 rpm for 5 min.
