MMP9 and MMP13 mRNA levels are down-regulated in response to SPDEF expression. RT-PCR was performed on a host of genes that have either previously been reported to be regulated by SPDEF or have a known role in prostate tumor progression and/or metastasis. MMP9 mRNA levels were significantly down-regulated in both the cell line and in the subcutaneous tumors overexpressing SPDEF. In addition, we found that MMP13 was also significantly down-regulated in the cell lines in vitro but not in the in vivo tumors. Expression measured was relative to housekeeping gene GAPDH. Graphs represent densitometry of three independent experiments of the RT-PCR data relative to the GAPDH.
