Immunization delivered by DCLV prime/DCLV boost in combination with PD-L1 blockade protects animals from recombinant vaccinia virus (vv) challenge. BALB/c mice were immunized with PBS, DCLV-Gag prime/DCLV-Gag boost alone, or DCLV-Gag prime/DCLV-Gag boost along with PD-L1 blockade. Two weeks following the boost immunization, all the groups were challenged with 107 PFU of vv-Gag. Vaccinia virus titers in paired ovaries harvested 7 days after challenge were determined on CV-1 cells. Titers from each mouse are shown as data points, and the mean value for each group is shown with a horizontal line. P values were calculated using one-way Anova followed by a Bonferroni nonparametric posttest (ns, not significant; *P < 0.05; **P < 0.01; ***P < 0.001).
