EGF treatment of cells before DsiRNA delivery enhances entry and silencing. (a) Well-differentiated airway epithelia from three donors were treated apically with EGF at a concentration of 100 µg/ml for 15 minutes before transfection with DsiRNAs against HPRT (250 nmol/l). RNAs were harvested for quantitation 24 hours later, as above. Mean levels (±SD) were calculated from three biological replicates (in triplicate) *P < 0.02, **P < 0.01 (Student's t-test). (b–c) Well-differentiated human airway epithelia were transfected with a formulation of DIG-HPRT DsiRNA and Transductin with EGF pretreatment (100 µg/ml for 15 minutes) (b) or mock treated (c).
