Inhibition of platelet adhesion to fibronectin by r-mojastin 1 and native mojastin disintegrins. A total of 100 μL of disintegrins at varying concentrations was added to 10 × 106 platelets and incubated for 1 h at 37°C prior to adding to fibronectin-coated wells. P-nitrophenyl phosphate (PNPP) is used as the substrate solution. The ability of platelet phosphatases to catalyze the hydrolysis of PNPP to p-nitrophenol (chromogenic product) was measured at 405 nm. The IC50s were 62.2 and 58.6 nM for r-mojastin 1 and native mojastin, respectively. The vertical bars represent the standard deviation. n = 3.
