Purified peripheral blood monocytes were treated with DMSO, AG1478, or PP2 for 1 h. Pretreated monocytes were then mock- or HCMV-infected (MOI 5) for 24 h and total RNA harvested for Affymetric gene array analysis. A P value of ≤0.05 was used as the criteria for statistically significant genes among replicates. Genes with an average 1.5-fold (A) up-regulation or (B) down-regulation were considered to be regulated by infection. Cellular genes dependent on EGFR or Src activity were determined by comparing the genes in HCMV-infected replicates with those that were down-regulated or up-regulated an average of 1.5-fold or greater in AG1478- or PP2-pretreated, HCMV-infected samples.
